Doc Sportello is een privédetective in het Los Angeles van de jaren zestig. Op een dag staat zijn ex voor de deur met het bericht dat de miljardair op wie zij verliefd is gekidnapt gaat worden. Sportello stort zich op de zaak en voor hij het weet raakt hij verwikkeld in een bizar web van motieven en krijgt hij te maken met surfers, oplichters, drugsverslaafden, een moordlustige woekeraar, een undercover saxofonist, en tot slot een mysterieuze entiteit die bekend staat als de Golden Fang.

Een mysterieuze vrouw ontsnapt aan een verkeersongeluk met een tas vol geld, maar zonder haar geheugen. Ondertussen is Betty Elms in L.A. verschenen, in de hoop een filmcarrière te kunnen beginnen.

FBI-agent Chet Desmond en Sam Stanley onderzoeken de moord op serveerster Teresa Banks. Op het bureau begint FBI-agent Dale Cooper ondertussen te geloven dat er iets gruwelijks gaande is en dat zijn mysterieuze dromen voorspellen wat er gaat gebeuren.

Chiyoko Fujiwara, ooit de grootste actrice van Japan, leidt een rustig, teruggetrokken leventje, ver weg van alle publiciteit. Toch stemt ze in met een interview met filmproducent Genya Tachibana. Wanneer ze hem vertelt over haar leven, beginnen feit en fictie met elkaar verstrengeld te raken.

De film draait om Dean Stiffle en alle inwoners van zijn omgeving in een buitenwijk. Als zijn beste vriend Troy zelfmoord pleegt worden al hun levens op de kop gezet, met gevolgen die een groot effect zullen achterlaten op iedereen.

Een belangrijk werk in de filmgeschiedenis. 'Zeer ongewoon, provocatief, heeft in geen enkele traditie een plaats', verklaarde Buñuel. De film is werkelijk zonder enige schijnbare samenhang en gaat in tegen alle logica; het perfekte voorbeeld van het 'automatische schrijven' dat geliefd is bij de surrealisten. We zien een nachtmerrie: een oog wordt door een scheermes doorkliefd, seminaristen trekken rond met piano's vol met ezels, die in staat van ontbinding verkeren. Dit meesterwerk van de vreemdheid en de fantasie is erg heftig en niet zonder symbolen, waarvan het belangrijkste het najagen van volledig nonconformisme lijkt te zijn, als reactie tegen de burgerlijke dwang. Aan het scenario is meegeschreven door Salvador Dalí.

Paul Hackett maakt een afspraakje met Marcy, maar op weg naar haar flat toe waait zijn geld uit het raampje van de taxi. Hierdoor is het voor hem onmogelijk om thuis te komen, en is hij genoodzaakt om de hele nacht door te brengen in SoHo, waar de vreemdste types hem in bizarre situaties doen belanden.

Rijke vrouwenversierder David ontmoet de mooie Sofia en valt als een blok voor haar. De volgende dag wordt hij door een ex-vriendinnetje opgehaald, die in een hysterische bui met de auto een ongeval veroorzaakt. David overleeft het ongeluk, maar zijn gezicht is vreselijk verminkt. Het geluk lijkt weer te keren wanneer David met Sofia herenigd wordt, en artsen zijn gezicht in de oude staat kunnen herstellen.

De Londense politie en Ahme, een hooggeplaatst lid van de cultus, die er niet meer in gelooft, proberen Ringo te helpen.
