Drive My Car berkisah tentang seorang pria bernama Yusuke Kafuku (Hidetoshi Nishijima) kehilangan istrinya secara mendadak. Dua tahun setelah kematian sang istri, ia memutuskan untuk melanjutkan karirnya sebagai aktor dan sutradara panggung teater. Pada suatu kesempatan, ia menerima tawaran untuk sebagai pengarah produksi Uncle Vanya di sebuah festival teater di Hiroshima.
