Den tamme papegøje Blu regnes for den sidste af sin art, men så får hans ejer nys om, at der findes en mage til Blu i Rio de Janeiro. Ved ankomsten til Brasilien når Blu knap nok at møde sin udkårne Jewel, før begge bliver kidnappet af fæle smuglere. Det lykkes de to araer at undslippe, men for at overleve i det fri må Blu lære at flyve - en kundskab, han aldrig har haft brug for i sit trygge bur.
The Rock spiller håndlangeren Beck. Han er manden, der bliver sendt ud som sidste mulighed, når der skal inddrives gæld, eller når uregerlige personer skal ind til en “lille snak med chefen”. Nu er fidusen bare, at den skruppelløse guldbaron Hatcher (Christopher Walken) mener, at han ejer hele junglen og alt, hvad der er i den. Der er også nogle oprørere, der render rundt i junglen og venter på at kunne smide Hatcher på porten. De vil også rigtig gerne have fingrene i skatten, så de kan finansiere deres oprør. Beck havner midt i denne konflikt, og det går ikke stille for sig.
Vi starter på Sydpolen hos en stakkels frysende pingvin, der drømmer om at komme nordpå til varmere himmelstrøg, men snart er vi i Andesbjergene med et flyvende æsel, og herfra går det hurtigt videre på en musikalsk rejse til Baia, Mexico City, Vera Cruz og Acapulco Beach.
