Tanjiro og vennerne har afsluttet deres rehabiliteringstræning på Butterfly Mansion og ankommer til deres næste mission ombord på Mugen-toget, hvor over 40 personer er forsvundet i løbet af meget kort tid. Tanjiro og søsteren Nezuko slutter sig sammen med deres venner Zenitsu og Inosuke til en af de mægtigste sværdkæmpere i Dæmonjægerkorpset - Flammesøjlen (“Flame Hashira”) Kyojuro Rengoku. Deres mission: at konfrontere dæmonen der gemmer sig i mørket ombord på Mugen-toget.
