اقتباساً عن سلسلة الكوميكس الشهيرة الرجل العنكبوت تدور أحداث الفيلم حول (بيتر باركر) الطالب اليتيم الهادئ الفقير الخجول الذي يتعرض في أحد المعارض العلمية لعضة عنكبوت معدل وراثياً مما يعطيه قدرات خارقة، بينما يحب بيتر (ماري) في صمت لأنها ترافق صديقه (هاري أوزبورن)، نجد أن (نورمان) والد هاري المليونير غريب الأطوار يستخدم نوعًا جديدًا من مخدرات تعزيز الأداء ليصبح العفريت الأخضر، فيصبح لزاما على بيتر مواجهته لإنقاذ عائلته وأصدقائه.
